Fucoidan with Antioxidant Effect on DPPH and Hydroxyl Radical Modulates Matrix Metalloproteinase-9 in HT1080 Cell Line

Fucoidan is a uniquely-structured sulfated polysaccharide found in the cell wall of several types of brown seaweeds that have recently attracted a lot attention due to its anti-tumor potential of enzymedigested fucoidan extract. The aim of this study was to investigate the effect of fucoidan on antioxidant activity and matrix metalloproteinase-9 (MMP-9) in human fibrosarcoma cells, HT1080 cells. First of all, the direct scavenging effect of fucoidan on reactive oxygen species (ROS) was investigated in vitro. Furthermore, the inhibitory effect of fucoidan on activity of MMP-9 was determined by gelatin zymography western blot analysis. In this study it was observed that fucoidan exhibited the highest inhibitory effect on DPPH radical and hydroxyl radical among ROS and blocked DNA oxidation induced by hydroxyl radical produced by fenton reaction. Moreover it specifically increased activity of MMP-9 but decreased MMP-9 expression via inhibition of ERK-1, a MAPK. These findings demonstrated that fucoidan with antioxidant activity can regulate the activity and expression MMP-9 in human fibrosarcoma cells.
